Transgenic Projects Services and Rates

The following information refers to a generation of plasmid-based and BAC DNA transgenic mice in a B6SJLF2, FVB or C57BL/6 background.

Services

• Final purification of plasmid-based transgenes or analysis of BAC DNA transgenes
• Pronuclear injection of enough embryos to produce 20-25 pups
• Wean and ear tag two-week-old pups; take tail biopsy for genotyping
• Transfer of founders to investigator

Expectations

• Typically, transgenesis rates of 10 to 25 percent are achieved resulting in the generation of two to seven transgenic founders, although results can vary significantly among constructs.
• If no founders are identified from the initial set of injections, a consultation will be requested. If the genotyping assay is determined to be accurate and adequate, a second set of injections will be performed.

Guarantees

• At least two transgenic founders or a maximum of 400 injected embryos are guaranteed.
• Generation of founders using special strains is not guaranteed
• Up to 400 embryos/construct will be injected with no guarantee of founders for projects in which the investigator opts to proceed with injections using suboptimal DNA
• Transgenic expression is not guaranteed

Costs

Costs include the following:

• Purchase of all animals used for the project
• Housing of the surrogate females
• Housing of pups for up to one week following weaning
• Additional fees:
  • Housing costs: daily animal care if genotyping exceeds more than one week after tails snips are provided
  • Use of other, non-routine strains for microinjection; includes the purchase and per diems of singly housed males for length of project
